Theraputic Effect of Nicrodil versus Bone Marrow Derived Mesenchymal Stem Cells on Unilateral Ureteric Obstruction in Adult Male Albino Rats (Histological and Biochemical study)

Aim: Chronic kidney disease (CKD) is an international health problem, in which renal fibrogenesis is the final target to treat. In our work we highlighted two new strategies; Nicorandil and Bone marrow Derived Mesenchymal Stem Cells (BM- MSCs) that have shown to be effective in reversing renal fibrosis induced by partial unilateral ureteric obstruction (PUUO). Materials and Methods: The current study included ninety six Male Albino rats divided randomly into 4 groups 24 rats per each as follow: Control group. Group II: PUUO where two-thirds of the left ureter entrenched in psoas muscle, Group III: same surgical procedure as group II for 7 days then rats received 15 mg/kg/day nicorandil once daily for 21 days and group IV: same surgical procedure in group II for 7 days then rats were injected intravenous by 3×106 of labeled MSCs then rats were left for 21 days. Blood and kidney tissues were collected for biochemical, histological and molecular analysis. Results: Both Nicrodil and BM-MSCs treatment groups could amolirate kidney damage evidence by inhibition of the elevation in MDA and the reduction in TAC caused by PUUO also there was significant reduction of TNF, TGF, IL6, collagen I and α-SMA in addition to improvement on histological examination. However, there was significant difference between BM-MSCs and Nicrodil treated groups. Conclusion: In conclusion, our results suggest that BM-MSCs and Nicorandil improves renal fibrosis progression through their anti-apoptotic, anti-inflammatory and antifibrotic effects in male albino rats subjected to PUUO with more effectiveness of BM-MSCs in comparison to nicrodil.
